Development and optimization of an extraction and purification method for 6-hydroxy-L-nicotine from the growth medium of Paenarthrobacter nicotinovorans, GI-UAIC-2022-08

Project summary:


   Alzheimer’s disease (AD) is the most typical form of dementia characterized by memory and cognitive loss. One hallmark pathology of AD is the loss of forebrain cholinergic neurons. Nicotinic acetylcholine receptors (nAChRs) are present in the cholinergic neurons indicating their involvement in cognitive functions such as learning and memory. Nicotine (NIC) is a well-known agonist of nAChRs and was reported to improve attention, learning, and memory. However, the cardiovascular and addictive side-effects of NIC have limited its therapeutic use in AD. Considering the positive effects on the central nervous system, NIC is increasingly seen as a scaffold for the development of new therapeutic agents of AD in the form of nicotinic derivatives that do not cause the side effects of its precursor. One such derivative is 6-hydroxy-L-nicotine (6HLN), the first nicotinic derivative that comes from the NIC degradation pathway of Paenarthrobacter nicotinovorans pAO1. 6HLN showed promising effects in in vivo studies, improving cognition and lowering oxidative stress, eluding NIC’s side effects. Currently, 6HLN can be obtained using a P. nicotinovorans-based biotechnology. However, the extraction and purification method for 6HLN from the culture medium leads to obtaining a low yield and to a product contaminated with other nicotinic derivatives. Therefore, the present project proposal aims to develop and optimize an extraction and purification method for 6HLN from the culture medium of a P. nicotinovorans strain.

General objective
The main scientific objective of this research project is to develop and optimize an efficient method for the extraction and purification of 6HLN from the culture medium of a strain of Paenarthrobacter nicotinovorans.
Activities in 2023:

Step 1
. Simplification of the culture medium for the P. nicotinovorans strain
Act 1.1. Purchase of reagents, supplies and equipment.
Act 1.2. P. nicotinovorans strain cultivation on different simple culture media.
Act 1.3. Yeast extract replacement with biotin solution
Act 1.4. Mineral solution recipe simplification

Activities in 2024:

Step 2
. Extraction method optimization for 6HLN extraction
Act 2.1. Extractions of 6HLN using several organic solvents at different pH values.
Act 2.2. Data analysis and literature review
Step 3. Extraction and purification of a large amount of 6HLN from the culture medium of P. nicotinovorans, previously grown in the bioreactor.
Act 3.1. Using preset conditions for the extraction of a large amount of 6HLN from the culture medium of P. nicotinovorans grown under bioreactor conditions
Step 4. Results dissemination
