Microorganismul Paenarthrobacter nicotinovorans (numit anterior Arthrobacter nicotinovorans) este unul din putinele organisme vii ce are capaciåtatea de a descompune alcaloidul nicotina si de a-l utiliza ca sursa de carbon si  nitrogen. Proprietate se datoreaza prezentei mega-plasmidului pAO1, entitate genetica independenta avand dimensiunea de 165 kB. pAO1 a fost complet secventiat si contine 2 grupe mari de gene: un prim grup implicat in metabolismul nicotinei si al doilea grup ce formeaza un operon implicat in metabolismul glucidelor. Este acceptat faptul ca genele implicate in catabolismul nicotinei au fost recrutate prin transfer orizontal de gene si grefate pe un plasmid ancestral, ce continea operonul implicat in metabolismul glucidelor (Igloi &  Brandsch 2003).

Principala tema de cercetare este organizarea moleculara si biologia megaplasmidul pAO1, in special in legatura cu mecanismele de protectie impotriva stresului oxidativ generat de metabolizarea nicotinei, metabolismul  glucidelor si aplicatiile biotehnologice ale acestor 2 cai. Utilizand o combinatie de tehnici bioinformatice am identificat substrate posibile pentru calea metabolica de degradare a glucidelor codificate de pAO1. Compusii selectati au fost apoi testati si am identificat ca substratul metabolic al celui de-al doilea grup de gene de pe pAO1 este xiloza.


Operonul codificand calea de degradare oxidativa a xilozei codificate de pAO1.


In ultimii ani prioritara a devenit evaluarea aplicatiilor medicale ale derivatilor nicotinici produsi de Paenarthrobacter nicotinovorans. Utilizand metode bioinformatice am testat potentialul derivatilor nicotinici de a se de receptorii nicotinici ai acetilcolinei. Am identificat astfel 6-Hydroxy-Nicotine (6HNic)  ca un potential ligand al receptorilor mentionati. In collaborare cu  dr. Lucian Hritcu am demonstrat ca intradevar 6HNic poate modula activitatea receptorilor nicotinici si imbunatati memoria intr-un model animal al bolii Alzheimer. In prezent, dezvoltam o metoda biotehnologica de producere a 6HNIC.

    Proiectele recente au ca scop studil integrat genomic, transcriptomic si proteomic al tulpinii Paenarthrobacter nicotinovorans pAO1 și a căii de degradare a nicotinei existente în aceast microorganism.

Pana acum, laboratorul coordonat a produs si facut publice urmatoarele seturi de date relevante:

  • primul genom complet si adnotat al microorganismului Paenarthrobacter nicotinovorans ATCC 49199: GenBank CP089293 si CP089294, precum si datele brute asociate PRJNA693273

  • un set de date de transcriptomica ce descriu metabolismul nicotinei in Paenarthrobacter nicotinovorans ATCC 49199: Gene Expression Omnibus GSE240220

  • un set de date de proteomica ce identifica proteinele exprimate in Paenarthrobacter nicotinovorans in prezenta nicotinei folosind nanoLCMSMS PXD008756

  • un set de date de proteomica ce descrie dinamica expresiei enzimelor implicate in metabolizarea nicotinei in Paenarthrobacter nicotinovorans - PXD012577
